I have laminate flooring on the main level of my house, and the upstairs bathroom sprung a leak and got all over the laminate on the main level.

How do I make sure that the laminate doesn't get damaged? Is there a way I can tell if it is already damaged? Because it sat wet all day while I was at work.

I'm no manufacturer (who would be the one who would need to tell you this) but if it's COMPLETELY dry and so is your sub-floor, and it still clicks back together than I'd say you're good. Watch for the tongue and groove being brittle (just in case it's low quality flooring which has broken down with the water). Most laminates these days are pretty good about water resistance though. Good luck!

So do I have to pull the flooring out in order to find out if the sub-floor is wet? How do I go about doing that?

Is there a way to tell if it needs replaced by just looking at it?

you should have swelling in the seams if water got into it. If not and the floor looks fine then I wouldnt worry about it. (if it aint broke then dont fix it)
